Within a certain concentration range, the measured absorbance value A595 is directly proportional to the protein concentration. The Bradford method for determining protein concentration is not affected by the chemical substances in the vast majority of samples. The concentration of mercaptoethanol in the sample can be as high as 1M, and that of dithiothreitol can be as high as 5mM. However, it is affected by slightly higher concentrations of detergents. It is necessary to ensure that the concentration of SDS is below 0.1% and that of Triton X-100 is below 0.1%. Tween 20, 60, 80 is less than 0.06%.
