Adenosine triphosphatase (ATPase) is a hydrolase and an enzyme that catalyzes ATP synthesis. ATPase can be divided into membrane ATPase, myosin ATPase and mitochondrial ATPase according to the activator, inhibitor and enzyme location. ATPase can hydrolyze adenosine triphosphate into adenosine phosphate and phosphoric acid, and this enzyme only acts on the high-energy bond between phosphoric acid and phosphoric acid, thus releasing a lot of energy. Its catalytic reaction is as follows: A-P-P+H2O → A-P-P+H3PO4+energy.
The principle of ATPase staining kit (calcium cobalt method) is that the hydrolysis of adenosine triphosphate by myoglobin ATPase is pH-dependent, and then the effects of different pH on ATPase activity are used to distinguish muscle fibers.
