Polylysine can be used to promote cell adhesion growth.Principle: Polylysine solution is a widely used adhesive for tissue sections and glass slides. The interaction between cationic molecules and anions on tissue sections produces strong adhesion. The properties of the surface of the culture bottle are crucial for cell culture. Glycoproteins (negative) on the cell surface are easily adsorbed on hydrophilic surfaces. Therefore, if there is a considerable amount of positive charge on the cell culture surface, it can promote cell adsorption more effectively. This is precisely the importance of using polylysine to optimize the cell culture surface. Application: Suitable for anti slip processing of glass slides used in histology, immunohistochemistry, frozen sections, cell smears, in situ hybridization, etc., to prevent tissue detachment during experimental procedures. It can also be used for cell culture to increase cell adhesion ability. Solution preparation: Dissolve in deionized water and prepare the mother liquor for later use. Store at -20 ℃. Glass slide processing: 1 Dilute polylysine solution with sterilized water, with a general usage concentration of 0.01%. two Soak the glass slide in diluted polylysine solution for 5 minutes. Note that increasing the time will not improve the wrapping effect. three Dry in a 60 ℃ oven for 1 hour, or dry overnight at room temperature of 18-26 ℃ for later use. Attention: 1 For every 100mL of diluted polylysine solution, 40-90 slides need to be coated. Exceeding 90 slides will affect their adhesion. two The previous glass slide must be kept clean. If necessary, clean with a 70% ethanol solution containing 1% HCl. three The released polylysine solution should be kept stable at 2-8 ℃ for at least 3 months. four Used diluent should be filtered, and if turbidity or bacterial growth occurs, it should be discarded The concentration of polylysine can be higher, and the absorbed substance can be reused at least 20 times. Cultivation bottle processing: 1 Coated with culture plates or bottles, the concentration of polylysine is generally 0.01%, 2 Prepare 100ml of triple distilled water and subject it to high-pressure sterilization treatment. three 0.01% polylysine was evenly applied to the surface of the culture substrate at a rate of 50 microliters per square centimeter, and allowed to stand at room temperature for 5 minutes to remove excess liquid. four Add sterile water, rinse repeatedly three times, and perform aseptic operation. five After processing, sterile air dry for later use. six Polylysine that remains sterile after recycling can be reused repeatedly.
