pU6-gRNA

Beyotime pU6-gRNA is a vector designed for constructing mammalian expression plasmids that express specific guide RNA (gRNA) based on CRISPR/Cas9 gene editing technology. Users only need to design and synthesize target-specific DNA oligos according to the instructions, then anneal them to form double-stranded DNA. Following the protocol, users can create a complete expression plasmid for the specific sgRNA used in CRISPR gene editing. After successfully constructing the plasmid, co-transfection with a Cas9 expression plasmid or dCas9 (dead Cas9) transcriptional activation or repression plasmid into mammalian cells enables the selection of gRNA-expressing positive cells using Zeocin resistance. Positive cells typically exhibit the expected gene editing or gene expression regulation of the target gene.
